The major goals of our research are to understand how cell-cell, cell-matrix adhesion and microenvironment remodelling promote tumour cell migration and invasion.

In this context, we analyse the role of Flotillins. These proteins are upregulated in numerous human invasive tumours and are associated with bad prognosis. We also study how P-cadherin, a protein that is upregulated in invasive breast carcinoma and skeletal muscle sarcoma, promotes collective cell invasion by converting biochemical cues onto mechanical forces.

Flotillins are involved in regulating cadherin-mediated intercellular adhesion (Bodin et al, Journal of Cell Science, 2014, 127, 5139–5147; Guillaume et al, Journal of Cell Science, 2013, 126, 5293-5304)

P-cadherin expression induces biochemical cues that control mechanical forces to allow collective cell migration (Plutoni et al, Journal of Cell Biology, 2016, 212, 199–217)

To answer these questions we use different models: cell lines (in 2D and 3D culture systems), to dissect the involved signalling pathways, and zebrafish, as an in vivo model system. We use cellular and molecular biology techniques. Particularly, we use fluorescent microscopy and video microscopy on live and fixed cells or zebrafish as well as biophysical approaches to study membrane dynamics and the forces generated at adhesion sites thanks to the imaging facility MRI and our collaborations.
