Montpellier Genomic Collections manages genomic libraries and offers a high-throughput cloning service.

Application portfolio:

  • High content screening (HCS) genome-wide,
  • Request of specific ORFs from human, yeast, mouse and Ciona intestinalis libraries for all application types,
  • Gene modification at low, medium and high throughput (CRISPR/Cas9 technology).

Main expertise/Services :

  • Libraries: the facility manages the ORF libraries of four species (human, mouse, yeast, and Ciona intestinalis). It has three human libraries: i) the version 5.1 includes 15483 ORF clones corresponding to 2794 genes; ii) the version 8 includes 12 230 clones (i.e., 11 149 non-redundant ORFs). The ORF clones include the coding sequence localized between the initiation codon and the stop codon, but without the 5’ and 3’ untranslated regions (5’ and 3’ UTR), thus allowing the addition of a tag in 3’; and iii) the ORFeome library (LifeTechnologies) that includes the stop codon.
  • Vector development: the facility manages and expands a library of destination vectors compatible with the Gateway cloning system.
  • High throughput cloning: we can isolate clones, create new clones in pDON vectors, and recombine ORF clones with different destination vectors.
  • Other services: plasmid DNA extraction (12, 48 and 96 samples), automated analysis by capillary electrophoresis.

MGC also develops the CRISPR/Cas9 technology. This system allows performing gene editing, knock out, and AAVS1 safe harbour targeting. MGC develops a range of vectors that can be easily used and has created a shared vector library. These vectors allows the C-terminal and N-terminal tagging of an endogenous gene of interest (GFP, HA, degron).
